Two dimensional HPLC facilitates the separation of complex mixtures by using different chromatography modes as sequential steps. This means that the output from the first HPLC column is automatically or manually transferred to the next HPLC column in order to observe and quantitate many more components of the analyte mixture.
There are many two possible two dimensional analysis systems. In traditional elution mode, one can use anion exchange in the first step and reverse phase in the second step (eAIXLC X eRPLC) or one can use reverse phase in the first step and a different reverse phase (with different selectivity )in the second step (eRPLC X eRP'LC); there are many other useful possibilities. Such two dimensional HPLC analysis systems enable separation of biomolecules that cannot be separated by any other means. Two dimensional HPLC analyses allow the observation of many more component peaks.
